Stem cells and the formation of the male germ line in the acoel Isodiametra pulchra

Abstract
Acoela are a group of acoelomate worms that have a pluripotent stem cell system (neoblasts). According to recent molecular phylogenetic studies, they are a sister group to the Bilateria. Recently, there has been growing interest in the study of neoblasts in Acoela. Given their exceptional function as the sole source for every cell type (both somatic and germ cells), it is important to know if there is a morphological difference between neoblasts and the male primordial germ cells. This difference can be detected at the ultrastructural level by either studying the morphology or by performing an immunogold labelling with antibodies against neoblast- or germcell specific proteins such as Vasa and Piwi. The process of forming mature spermatozoa out of primordial germ cells or spermatogonia is called spermatogenesis. For general morphological descriptions, it is important to define the stages of spermatogenesis. Besides knowing which cells are neoblasts and which cells are early stages of spermatogenesis, it is pivotal to understand how these cells are organized within the testes, as the spatial organisation of the testes in acoels varies greatly. The objectives of this study are two-fold: describing the stages of spermatogenesis in I. pulchra at the ultrastructural level and defining the spatial organization in the testes.
