Project: Studies on TGF isoforms and receptors and their role in Treg-dependent immuno-suppression and remodelling in chronic rhinosinusitis (CRSwNP). Impact of Staphylococcus aureus superantigens

1) Identification and quantification of T-cell subgroups including Tregs (CD4-CD25+, FOXP3+) in chronic sinusitis, using tissue and PBMCs, in relation to TGF-B activation, TGF-B isoform and TGF-B receptor expression in the tissue and T-cells by immunohistochemistry and real time PCR. 2) Quantification of key cytokines (IL-10, TGF-B, IL-4, IL-5, IFN-gamma) and ECM/metalloproteases/TIMP in the same tissues/locations by Cytometric beat assay (CBA)/ELISA. These data will lead to a detailed knowledge on presence and function of Tregs and their relation to the inflammatory and remodelling factor milieus in sinus disease, and within different compartments of disease manifestation. Functional assays on the impact of SAEs on TGF-B isoform expression in Tregs (FOXP3+), Treg precursors and other TGF-contributing cells (e.g. macrophages).
