Project: Effect of superantigens on inflammatory and structural cells in upper respiratory airway diseases.

- In this project we aim to study the role of the S. aureus enterotoxins in inflammatory and structural cells of the respiratory mucosa, using nasal polyposis (NP) and chronic rhinosinusitis (CRS) as disease models. As samples we will use peripheral blood leucocytes (PBLs), and nasal tissue fibroblasts from patients with NP, CRS and control group. Cells will be cultured and stimulated with S. aureus enterotoxins in different conditions. Furthermore, cells and supernatants will be assayed for qRT-PCR, ELISA, and ImmunoCAP to study the changes in AA biosynthetic pathway and alterations in eosinophilic inflammatory markers. Effect of SAEs on remodelling will be functionally tested in tissue fibroblasts in a wound-healing test.
