These nanoplastics display a complex chemical and physical composition, broad dispersity and poorly understood effects on the function of the mammalian gastrointestinal tract. To address these knowledge gaps, tractable models together with a selection of the most appropriate (bio)analytical methods, enabling for live multi-parameter analysis with single cell resolution, are much needed.
Here we report a new method to study interactions of nanoplastics with the gut epithelium, based on combining adult stem cell-derived small intestinal organoid cultures with multi-parameter live Fluorescence Lifetime Imaging Microscopy (FLIM). Normally, gut organoids display a ‘basal-out’ topology with an apical membrane that is difficult to access. To address this, we optimized the conversion of porcine and mouse small intestinal organoid cultures to an ‘apical-out’ topology (confirmed by a phalloidin staining), which improves apical-membrane availability without disturbing a 3D integrity of organoids and facilitates studying epithelial interactions with drugs, microbiota and nanoplastics. Live ‘apical-out’ organoids were exposed to a set of nanoparticles exhibiting different surface charges, sizes of <200 nm and doped with red fluorescent dyes. In contrast to previous work with basal-out gut organoids, we observed an efficient accumulation of the nanoparticles from the apical side over the course of 24 h under regular growth conditions. We also compared staining of basal- and apical-out gut organoids with several fluorescent FLIM probes, enabling visualization of cell redox, mitochondrial membrane potential and lipid droplets. Interestingly, a more efficient uptake was observed for many dye structures including the lipid-sensing Nile Red and the membrane tension-sensing Flipper-TR probes.
Together with segmentation of stem cells with tracers such as Lgr5-GFP, labeling cell redox and ‘apical-to-basal’ membrane transport, the presented methodology enables to address the effects of nanoplastics on stem cell and enterocyte function, their metabolism and efficiency of nutrient absorption, in a live and multi-parameter setting.

Moreover, increased evidence suggests that autism spectrum may manifest differently in males and females which could lead to a diagnostic sex/gender bias. To fully understand the nature of autism spectrum, it is important to produce scientific information of autism spectrum in both sexes/genders. The aim of this study was to examine speech disfluencies in autistic and non-autistic young adults, and to compare disfluencies between males and females.
Methods: The participant groups consisted of autistic males (n = 7), autistic females (n = 7), non-autistic males (n = 7), and non-autistic females (n = 7) aged 19–30 years. There were no group differences in age, general ability, and verbal comprehension. Speech disfluencies were analyzed from a narrative speech sample and categorized as typical, stuttering-like, and atypical disfluencies, which further formed the total disfluency frequency (e.g., Scaler Scott et al., 2014). Non-parametric statistical tests were used for group comparisons.
Results: Preliminary results showed that on average, males had higher frequency of each disfluency types when compared to females in both groups, however, statistically significant between-group differences were not detected. The autistic males produced significantly more typical and stuttering-like disfluencies than the non-autistic males or females. The autistic females produced significantly more stuttering-like disfluencies than the non-autistic females, but no other differences were observed. There were no significant differences in disfluencies between autistic females and non-autistic males.
Discussion: The mean disfluency profiles of the groups indicate a continuum in which the non-autistic females locate in the one end having the lowest disfluency frequency, and the autistic males locate in the other end having the highest disfluency frequency. The autistic females and the non-autistic males locate somewhere between those ends. The results support the conception that autism spectrum could manifest differently in males and females.

The key role of these technologies in the development of a more sustainable future implies the need for the evaluation and monitoring of both supply risks as well as environmental and social impacts of a number of raw materials in the supply chain. These raw materials that are important to the economy and might be under supply risk are referred to as Critical Raw Materials (CRMs) in the EU. The integration of Life Cycle Sustainability Assessment (LCSA) - well established for sustainability evaluation - and Criticality Assessment (CA) – increasingly used as governance tool - is therefore consequent to support decision-making regarding efficient use those natural resources.
Based on a critical review of CA methods within and outside the framework of an LCSA, this research aimed to investigate the compatibility of CA methods with the life-cycle approach. The methods range from specific CA methodologies (e.g., NRC (USA) and EC-CA (EU)) to the existing methods integrating CA and LCSA (e.g., ESSENZ and GeoPolRisk). The evaluation of the methods was based on a set of criteria (e.g., acceptance and credibility) and further analysis of compatibility with frameworks from ISO 14040-44 and UNEP-SETAC. The current challenges for integration in the field are identified, namely: interpretation of criticality within the three pillars of sustainability (social, economic or environmental); the incompatibility among inventories and the characterization of material’s criticality; arbitrariness in the interpretation of what is “critical”; and the uncertainty intrinsic to CA models. Potential solutions towards the operationalization of criticality indicators in a product-oriented LCSA include the definition of the impact pathway of criticality in LCSA, the linkage of criticality indicators to product/technological flows, the use of intermediate indicators (supply risk and economic importance), the characterization of criticality at the normalization and weighting step, and addressing uncertainties in an LCSA. A 12-year old girl was hospitalized in a peripheral hospital. She had been suffering from muscle pains and edema for over a month, that had started after an episode of fever and throat pain. At admission, the diagnosis of viral myositis was made. She was transferred to the pediatric intensive care unit of Ghent University Hospital. Juvenile dermatomyositis was suspected, and high dose corticosteroids were initiated.
A week after admission, she developed abdominal pain and distension combined with respiratory deterioration. RX thorax showed a significant amount of intraperitoneal air, so a CT abdomen was performed. This showed significant free air and fluid, caused by an intestinal perforation. Exploratory laparoscopy revealed a large perforation of the duodenum which was sutured laparoscopically and combined with an omentoplasty.
A week after surgery, a fall in hemoglobin level from 8.1 to 5.0 g/dL was noticed. Moreover, blood was aspirated from the nasogastric tube and melena was present. Gastroscopy confirmed a large duodenal ulcer that was treated with gel application. Control gastroscopies in the week afterwards showed positive evolution of the ulcer. However, two days after the third gastroscopy, her abdomen started to become tender and distended. Gastroscopy and CT scan revealed a recurrence of the duodenal bulb perforation.
An exploratory laparotomy was performed. This revealed a contaminated abdomen with biliary peritonitis. A perforation at the earlier suture was identified, but it had become more of a laceration and extended into the descending part of the duodenum. A Billroth 2 resection was deemed unfeasible, so a Whipple resection was performed. No reconstructions were made immediately and both pancreas and biliary tract were derived externally. Anastomoses could be performed two days later.
The patient was weaned from sedation and ventilation two weeks later. Peroral feeding was initiated a month after surgery. She was transferred to a regular pediatric unit two months after initial admission and is currently doing well.
Conclusion:
Juvenile dermatomyositis with gastrointestinal perforation is extremely rare and lethal according to literature.
